Potato dextrose media i.e PDA is used for the cultivation of fungi. Potato dextrose is a general media.which is recommended for plate count methods for foods.
It encourages mold sporulation and pigment production in dermatophytes.
Agar is used as solidifying agents.
Procedure
1. Boil 200g sliced, unpeeled potatoes in 1 litre distilled water for 30 min.
2. Filter through cheese cloth, is a potato infusion.
3. Mixed with glucose , peptones, yeast extract, potato starch.
4. Autoclave 2 hr at 121 degree celsius.
5. Dispense 20-21 ml portions into steril petri dishes.
6. Final PH 5.6 ± 0.2
